Naegleria fowleri is a free-living, unicellular amoeba that can be found in warm, freshwater environments such as lakes, rivers, and hot springs. While it is generally harmless, Naegleria fowleri can cause a rare but highly fatal disease known as naegleriasis, also called primary amoebic meningoencephalitis (PAM). PAM is an infection of the brain and spinal cord that can lead to severe brain damage and death within just a few days.

One of the key proteins of Naegleria fowleri is its membrane protein, which plays a critical role in the amoeba’s interactions with its environment and host. The membrane protein is a large, complex molecule that spans the membrane of the amoeba and is exposed on the surface of the cell.

The membrane protein of Naegleria fowleri is highly immunogenic, meaning that it is capable of inducing a strong immune response in humans and animals. This has made it a target for the development of diagnostic tests and vaccines for naegleriasis.

Researchers are also studying the functions of the membrane protein to gain a better understanding of how Naegleria fowleri interacts with its environment and host. This could lead to the development of new treatments for naegleriasis, as well as other diseases caused by related amoebas.

In addition to its membrane protein, Naegleria fowleri also produces other important proteins, such as enzymes and virulence factors. These molecules play a critical role in the pathogenesis of naegleriasis and are also being studied for their potential diagnostic and therapeutic applications.

Naegleria fowleri cDNA and recombinant antigen can be used for the development of diagnostic tests for the detection of Naegleria fowleri infection. The cDNA can be used for the development of PCR-based assays, while the recombinant antigen can be used for the development of ELISA-based assays. These assays can be used to detect the presence of the organism in samples such as cerebrospinal fluid, or to detect antibodies to the organism in serum samples. These assays can be used to diagnose Naegleria fowleri infection in humans and animals, thus facilitating early diagnosis and treatment.

The use of recombinant proteins/cDNA in academic research and therapeutic applications has skyrocketed. However, in heterologous expression systems, successful recombinant protein expression is dependent on a variety of factors, including codon preference, RNA secondary structure, and GC content. When compared to pre-optimization, more and more experimental results demonstrated that the expression level was dramatically increased, ranging from two to hundred times depending on the gene. Bioclone has created a proprietary technology platform that has resulted in the creation of over 6,000 artificially synthesized codon-optimized cDNA clones (cloned in E. coli expression Vector), which are ready for production of the recombinant proteins.
