Giardia lamblia cDNA and recombinant antigen
Giardia lamblia, also known as Giardia intestinalis, is a flagellated intestinal parasite that infects humans and animals. It causes a diarrheal disease called giardiasis, which is prevalent in developing countries and is a significant cause of morbidity and mortality in children. This article aims to provide an overview of various surface proteins, antigens, and other biomolecules produced by Giardia lamblia that contribute to its pathogenicity.
Variant-Specific Surface Proteins (VSPs): The VSPs are a family of variable surface proteins that contribute to antigenic variation in Giardia lamblia. These proteins are expressed on the surface of the trophozoites and play a crucial role in immune evasion by the parasite. The VSP family includes several proteins, such as R1 and M165, and specific surface proteins like M30, M20, M21, M21-1, H7-1, H7, 1267, VSP136b, and VSP4A1.
Trophozoite Surface Protein 11 and 417 deltas: Trophozoite surface proteins 11 and 417 delta are two critical biomolecules expressed on the surface of Giardia lamblia trophozoites. These proteins are involved in host-parasite interactions and contribute to the pathogenicity of the parasite.
Labeled Trophozoite Antigen 417: The labeled trophozoite antigen 417 is a highly specific antigen produced by Giardia lamblia. It is commonly used in diagnostic tests to detect the presence of the parasite in stool samples.
Proliferating Cell Nuclear Antigen (PCNA): PCNA is a highly conserved protein expressed by many eukaryotic cells. In Giardia lamblia, PCNA is involved in DNA replication and repair and plays a critical role in the survival of the parasite.

Liver Stage Antigen-Like Protein (LSA): LSA is a recently discovered protein expressed by Giardia lamblia that shares significant homology with Plasmodium falciparum’s liver stage antigen-1. LSA is considered a promising target for the development of a Giardia lamblia vaccine.
Minor Histocompatibility Antigen H13: H13 is a minor histocompatibility antigen expressed by Giardia lamblia that has been shown to induce protective immunity against the parasite in animal models.
Serologically Defined Colon Cancer Antigen 1: Serologically defined colon cancer antigen 1 is a protein expressed by Giardia lamblia that shares structural homology with the human colon cancer antigen CO17-1A. This protein is considered a potential target for the development of an anti-giardiasis vaccine.
The use of recombinant proteins/cDNA in academic research and therapeutic applications has skyrocketed. However, in heterologous expression systems, successful recombinant protein expression is dependent on a variety of factors, including codon preference, RNA secondary structure, and GC content. When compared to pre-optimization, more and more experimental results demonstrated that the expression level was dramatically increased, ranging from two to hundred times depending on the gene.
