Eimeria tenella cDNA and recombinant antigen
Eimeria tenella is a common coccidian parasite that causes avian coccidiosis, a significant disease in poultry production worldwide. This parasite has evolved a complex lifecycle that involves multiple developmental stages within the host’s intestinal tract. The parasite’s success in colonizing the host relies on its ability to evade the host’s immune system, which is facilitated by several proteins, including the surface antigen and profilin.
The surface antigen of Eimeria tenella plays a critical role in the parasite’s lifecycle by helping the parasite to evade the host’s immune system. The antigen is a type of glycoprotein that is expressed on the surface of the sporozoites and merozoites. The surface antigen is responsible for initiating an immune response in the host, leading to the production of specific antibodies that recognize and bind to the parasite. However, Eimeria tenella has developed strategies to avoid this response by regularly changing its surface antigen. This allows the parasite to evade the host’s immune system and continue to cause damage.
Profilin is a small protein found in Eimeria tenella that plays a crucial role in the parasite’s lifecycle. The protein is involved in the formation of actin filaments, which are essential for the parasite’s motility and invasion of the host’s intestinal cells. Profilin acts as an important regulatory protein, controlling the polymerization and depolymerization of actin filaments. By regulating the actin filaments, profilin enables the parasite to move within the host’s intestinal cells and evade the host’s immune system.
The surface antigen and profilin in Eimeria tenella play critical roles in the parasite’s ability to cause disease in the host. The surface antigen helps the parasite to evade the host’s immune system by continually changing its surface structure, while profilin regulates the formation of actin filaments, enabling the parasite to move within the host’s intestinal cells. Together, these proteins enable Eimeria tenella to colonize the host’s intestinal tract and cause significant tissue damage and hemorrhage.
The use of recombinant proteins/cDNA in academic research and therapeutic applications has skyrocketed. However, in heterologous expression systems, successful recombinant protein expression is dependent on a variety of factors, including codon preference, RNA secondary structure, and GC content. When compared to pre-optimization, more and more experimental results demonstrated that the expression level was dramatically increased, ranging from two to hundred times depending on the gene.
