Porphyromonas gingivalis is a gram-negative bacterium that is commonly associated with chronic periodontitis. The bacterium expresses a variety of virulence factors, including numerous immunoreactive antigens, that contribute to its pathogenicity. Among these are the 50 kD antigen PG1, the 89 kD antigen PG87, the PgaA protein (also known as Antigen PgaA), the 36 kDa antigen PG14, the 46 kDa antigen PG99, the 47 kD antigen PG120, the 47 kDa antigen PG97, the 52 kD antigen PG41, the 53 kD antigen PG123, the 61 kDa antigen PG91, the 63 kDa antigen PG102, the 84 kDa antigen PG93, and the 92 kDa antigen PG21. Additionally, the bacterium expresses receptor antigens, including RagA and B, and the immunoreactive 23 kDa, 32 kDa, and 46 kDa antigens. These antigens play important roles in the bacterium’s ability to colonize and cause disease in the oral cavity.

50 kD Antigen PG1

The 61 kDa antigen PG91 is also known as the major fimbrial antigen, as it is a key component of the fimbriae, which are hair-like appendages on the surface of P. gingivalis. Fimbriae are important for the bacterium’s ability to adhere to host tissues and form biofilms, which are complex communities of bacteria that are resistant to immune responses and antibiotics.

The use of recombinant proteins/cDNA in academic research and therapeutic applications has skyrocketed. However, in heterologous expression systems, successful recombinant protein expression is dependent on a variety of factors, including codon preference, RNA secondary structure, and GC content. When compared to pre-optimization, more and more experimental results demonstrated that the expression level was dramatically increased, ranging from two to hundred times depending on the gene. Porphyromonas gingivalis cDNA and recombinant antigens can be used in a variety of applications, such as in vaccine development, diagnostics, therapeutic interventions and research. Vaccines developed using P. gingivalis cDNA and recombinant antigens may be used to prevent and/or reduce levels of periodontitis, a progressive and destructive gum disease caused by P. gingivalis. Diagnostic applications using these antigens could be used to detect and identify P. gingivalis infection in patients. Therapeutic interventions using P. gingivalis cDNA and recombinant antigens could be used to treat periodontitis, as well as other diseases caused by P. gingivalis. Finally, these antigens can also be used in research applications to further understand the molecular and cellular mechanisms of P. gingivalis infection.
