Mycobacterium bovis is a species of bacteria that is a member of the Mycobacteriaceae family. It is a slow-growing, acid-fast, and gram-positive organism that is the causative agent of bovine tuberculosis. Mycobacterium bovis typically infects animals, although humans can also become infected if they are exposed to infected animals, their products, or the environment. The most common form of transmission is through consumption of unpasteurized milk and milk products. Treatment of M. bovis infection involves a combination of antibiotics and other drugs. This bacterium produces several key antigens and proteins associated with this pathogen, including:

14 kDa and 16 kDa antigens: These are proteins produced by Mycobacterium bovis that are believed to play a role in the immune response of the host against the bacterium.

HSP 16.3: This is a heat shock protein produced by Mycobacterium bovis that is involved in the stress response and survival of the bacterium within the host.

19 kDa lipoprotein antigen lpqH: This is a cell surface lipoprotein produced by Mycobacterium bovis that has been implicated in the virulence of the bacterium, as it is required for its survival within host macrophages.

22 kDa protein: This is a protein produced by Mycobacterium bovis that is believed to be involved in the immune response against the bacterium.

27 kDa antigen Cfp30B: This is a protein antigen produced by Mycobacterium bovis that has been shown to induce an immune response in the host.

9.5 kDa culture filtrate antigen cfp: This is a secreted protein antigen produced by Mycobacterium bovis that has been implicated in the immune response against the bacterium.

Cell surface protein pirG: This is a protein produced by Mycobacterium bovis that is believed to be involved in the interaction between the bacterium and host cells.

16 kDaimmunoprotective extracellular protein: This is a protein produced by Mycobacterium bovis that has been shown to have immunoprotective properties, meaning it can help protect the host against the bacterium.

Immunogenic protein MPB64 antigen: This is a protein produced by Mycobacterium bovis that is known to be immunogenic, meaning it can induce an immune response in the host.

Lipoprotein lprG: This is a cell surface lipoprotein produced by Mycobacterium bovis that has been implicated in the virulence of the bacterium.

27 kDa lipoprotein antigen P27: This is a cell surface lipoprotein produced by Mycobacterium bovis that has been shown to be involved in the immune response against the bacterium.

Protein antigen 6 cfp6: This is a secreted protein antigen produced by Mycobacterium bovis that has been implicated in the immune response against the bacterium.

Secreted antigen 85-C fbpC 85C: This is a secreted protein antigen produced by Mycobacterium bovis that has been shown to be involved in the biosynthesis of mycolic acids, which are important components of the mycobacterial cell wall.

40 kDa antigen: This is a protein produced by Mycobacterium bovis that is believed to play a role in the immune response against the bacterium.

Mpb51 antigen protein FbpD: This is a protein produced by Mycobacterium bovis that has been implicated in the virulence of the bacterium.

Soluble secreted antigen Mpb53: This is a secreted protein antigen produced by Mycobacterium bovis that has been shown to be involved in the immune response against the bacterium.

WAG22 antigen: This is a protein produced by Mycobacterium bovis that is believed to be involved in the immune response against the bacterium.

The use of recombinant proteins/cDNA in academic research and therapeutic applications has skyrocketed. However, in heterologous expression systems, successful recombinant protein expression is dependent on a variety of factors, including codon preference, RNA secondary structure, and GC content. When compared to pre-optimization, more and more experimental results demonstrated that the expression level was dramatically increased, ranging from two to hundred times depending on the gene. Bioclone has created a proprietary technology platform that has resulted in the creation of over 6,000 artificially synthesized codon-optimized cDNA clones (cloned in E. coli expression Vector), which are ready for production of the recombinant proteins.

The cDNA can be used to identify the presence of the M. bovis genome in a sample, while the recombinant antigens can be used to detect antibodies against the pathogen. Through a combination of these techniques, a more accurate diagnosis of bovine tuberculosis can be obtained. cDNA can be used in polymerase chain reaction (PCR) techniques to detect the presence of M. bovis in a sample. The recombinant antigens can be used in an enzyme-linked immunosorbent assay (ELISA) to detect antibodies against the pathogen. Through these techniques, a more accurate diagnosis of bovine tuberculosis can be obtained.
