Leptospira interrogans cDNA and recombinant antigen
Leptospira interrogans is a spirochete bacterium responsible for causing leptospirosis, a zoonotic disease that affects both humans and animals. The detection and identification of this bacterium can be challenging due to its complex antigenic makeup. However, recent studies have identified several key antigens that may serve as potential biomarkers for improved pathogen detection.
Flagellar Filament 35 kDa Core Protein Antigen
The flagellar filament 35 kDa core protein antigen is a structural component of the bacterial flagellum that is exposed to the host’s immune system. This protein has been shown to induce an immune response in infected hosts, making it a potential biomarker for the detection of Leptospira interrogans.
Lipoprotein
Lipoproteins are a group of proteins that are covalently linked to lipids and are often found on the surface of bacterial cells. In Leptospira interrogans, several lipoproteins have been identified as potential biomarkers for pathogen detection. These lipoproteins can elicit an immune response in the host and are conserved across different strains of the bacterium, making them reliable targets for the development of diagnostic assays.
Surface Antigen Copenhageni LipL32
Copenhageni LipL32 is a surface antigen of Leptospira interrogans that has been shown to be highly immunogenic. This protein is expressed in all pathogenic strains of Leptospira interrogans and is therefore a reliable target for the detection of this bacterium.
Preprotein Translocase Subunit SecY
The preprotein translocase subunit SecY is a component of the bacterial protein translocation machinery. This protein has been identified as a potential biomarker for the detection of Leptospira interrogans due to its presence on the bacterial surface and its immunogenicity.
The identification of these key antigens of Leptospira interrogans has the potential to improve the accuracy and reliability of pathogen detection. The flagellar filament 35 kDa core protein antigen, lipoproteins, surface antigen Copenhageni LipL32, and preprotein translocase subunit SecY are all promising candidates for the development of diagnostic assays that can detect the presence of Leptospira interrogans in infected hosts. The validation and implementation of these biomarkers could lead to earlier diagnosis and treatment of leptospirosis, ultimately improving patient outcomes.
The use of recombinant proteins/cDNA in academic research and therapeutic applications has skyrocketed. However, in heterologous expression systems, successful recombinant protein expression is dependent on a variety of factors, including codon preference, RNA secondary structure, and GC content. When compared to pre-optimization, more and more experimental results demonstrated that the expression level was dramatically increased, ranging from two to hundred times depending on the gene. Bioclone has created a proprietary technology platform that has resulted in the creation of over 6,000 artificially synthesized codon-optimized cDNA clones (cloned in E. coli expression Vector), which are ready for production of the recombinant proteins.
Leptospira interrogans cDNA and recombinant antigen can be used in a variety of applications, including diagnostic and therapeutic applications.
1. Diagnostics: cDNA and recombinant antigen can be used in a variety of diagnostic tests, including nucleic acid amplification tests (NAATs), immunological assays, and PCR-based assays. These tests can be used to detect the presence and/or titer of Leptospira interrogans in a sample.
2. Vaccines: cDNA and recombinant antigen can be used in the development of vaccines against Leptospira interrogans. The recombinant antigens can be used to induce an immune response in individuals, which can protect them from infection with the pathogen.
3. Therapeutics: cDNA and recombinant antigen can be used in the development of therapeutics to treat individuals with leptospirosis. The recombinant antigens can be used to stimulate the immune system to target the bacteria, potentially reducing infection and disease severity.
