Mycobacterium tuberculosis is the causative agent of tuberculosis (TB), a serious infectious disease that affects millions of people worldwide. The bacterium possesses various antigens that play important roles in its pathogenesis and immune response. Let’s explore some of these antigens:

10 kDa chaperonin-protein: Also known as the heat shock protein 10 (Hsp10), this antigen is a chaperonin that is involved in protein folding and stress response. It has been shown to interact with the host immune system and modulate the immune response during M. tuberculosis infection.

45-47 kDa antigen: This antigen is a cell wall-associated protein of M. tuberculosis that is recognized by the host immune system during infection. It has been shown to elicit strong immune responses and is often used in diagnostic tests for TB.

AhpD and AhpC: AhpD and AhpC are two antioxidant enzymes of M. tuberculosis that are involved in protecting the bacterium from oxidative stress. They play a role in the ability of M. tuberculosis to survive and persist in the host immune response.

Antigen MTB48: Antigen MTB48 is a secreted protein of M. tuberculosis that is involved in the host-pathogen interaction. It has been shown to stimulate immune responses in infected individuals and is considered to be an important antigen in the context of TB.

The use of recombinant proteins/cDNA in academic research and therapeutic applications has skyrocketed. However, in heterologous expression systems, successful recombinant protein expression is dependent on a variety of factors, including codon preference, RNA secondary structure, and GC content. When compared to pre-optimization, more and more experimental results demonstrated that the expression level was dramatically increased, ranging from two to hundred times depending on the gene.
