Ehrlichia ruminantium cDNA and recombinant antigen
Ehrlichia ruminantium is a gram-negative bacterium that is the causative agent of heartwater disease in ruminants, which can lead to significant economic losses in livestock industries. As you mentioned, this bacterium has several proteins that act as antigens, including Immunodominant protein, Major antigenic protein 1, protein 1-2, MAP1, MAP1-1, and peroxiredoxin.
Immunodominant protein is a surface-exposed protein that elicits a strong antibody response in infected animals. It has been extensively studied and is considered an important diagnostic antigen for heartwater disease.
Major antigenic protein 1 (MAP1) is another surface-exposed protein that is highly immunogenic and has been identified as a potential vaccine candidate for heartwater disease.
Protein 1-2 and MAP1-1 are also surface-exposed proteins that have been shown to elicit an antibody response in infected animals.
Peroxiredoxin is an antioxidant enzyme that is involved in protecting the bacterium from oxidative stress. It has been identified as a potential diagnostic antigen for heartwater disease.
The use of recombinant proteins/cDNA in academic research and therapeutic applications has skyrocketed. However, in heterologous expression systems, successful recombinant protein expression is dependent on a variety of factors, including codon preference, RNA secondary structure, and GC content. When compared to pre-optimization, more and more experimental results demonstrated that the expression level was dramatically increased, ranging from two to hundred times depending on the gene. Bioclone has created a proprietary technology platform that has resulted in the creation of over 6,000 artificially synthesized codon-optimized cDNA clones (cloned in E. coli expression Vector), which are ready for production of the recombinant proteins.
The application of Ehrlichia ruminantium cDNA and recombinant antigen can be used in the development of diagnostic tests for the detection of Ehrlichia infections in ruminant animals. Diagnostic tests based on the detection of Ehrlichia ruminantium cDNA and recombinant antigen can also be used to differentiate between Ehrlichia-infected animals and those that are not infected. Furthermore, the cDNA and recombinant antigen can be used to develop vaccine candidates for the prevention of Ehrlichia infections in ruminants.
